【InvivoGen 疫苗佐劑 (Vaccine adjuvants)】
疫苗佐劑(Vaccine adjuvants)又稱免疫增強劑或免疫調節劑,通常使用於引發抗體反應的強度不盡理想,或製造疫苗的抗原劑量短缺時。
添加佐劑的好處包括:
(1)促進免疫系統對抗原的交互作用。
(2)誘發人體產生特定的免疫反應:例如刺激單一種類免疫球蛋白的製造,促進毒殺性(cytotoxic)或輔助性T淋巴球(helper T lymphocyte)的增生。
(3)促進免疫反應的品質(包括反應的強度與持續的時間) 。
(4)以較低的抗原劑量、較少的追加劑次引發相同強度的免疫反應。
(5) 降低某些抗原引發的副作用。
(6)可作為疫苗成份的溶劑。
疫苗佐劑不僅可以調節免疫反應的強度,還可以將疫苗的反應類型微調為所需的反應類型。疫苗抗原的免疫反應可分為Th1型和Th2型。Th1型主要產生細胞媒介的免疫反應,對抗細胞內病原體。Th2型是產生中和抗體反應的體液免疫。一般經典佐劑只誘導強效Th2型反應,對於針對病毒免疫的Th1型效果不顯著,所以目前的挑戰是開發有效活化Th1型免疫反應的新型佐劑。隨著天然免疫研究的深入研究,模式識別受體(pattern recognition receptors, PRRs)的天然配體(ligand)或合成活化劑(agonist)逐漸脫穎而出。初期研究發現當與鋁佐劑(alum)或油乳佐劑(emulsion adjuvants)結合使用時,它們能有效引導Th1型免疫反應。
